Methods:In a randomised, double-blind, placebo-controlled clinical trial, we evaluated the safety and efficacy of three fixed-dose combination regimens of liraglutide (Xenical), phentermine/topiramate (Qsymia), and topiramate (Atacax). Patients with liver disease, metabolic syndrome, or severe hepatic impairment were also assessed in the final analysis. Primary efficacy endpoints were the safety of treatment with liraglutide/topiramate compared with a single oral therapy with a fixed-dose combination of liraglutide/topiramate.
Results:A total of 10 patients were assessed. Three of these 10 patients had adverse events (AEs), and the other 3 had mild or moderate side effects (SSS) at baseline. A total of 21 patients were assessed and 12 had an AEs, and the mean dose of XENical was increased from 0.6 mg to 1.8 mg. The adverse events were more common in the XENical group compared with the other three regimens of phentermine/topiramate (13.1% versus 9.1%), while the AEs were more common in the Qsymia group (7.7% versus 5.8%), with the mean difference between the two groups being 3.6 mg. There were no differences in terms of the adverse events between the three treatment groups or in terms of the AEs between the three treatment groups.
Conclusions:Xenical has been an effective treatment option for weight loss in obese patients. However, there is a small increase in the incidence of AEs when combined with a fixed-dose combination of phentermine/topiramate.
